coli 의 identification을 위하여 gram 염색을 실시하였다. E.coli는 gram negative 균으로 peptidoglycan layer가 단일층으로 존재하고, 세포벽이 지방성분이 풍부한 lipopolysaccharide (LPS)로 이루어져 있어 organic decolorizer(탈색제)에 의해 crystal violet이 쉽게 탈색되어 따라서 염색액인 safranin O로 염색되어 붉은색을 나타낸다.
