In synthetic luminol process we need luminol’s precursor. That is
5- nitrophthalhydrazide. It is synthesized by the reaction of 3-nitrophthalic acid
with hydrazine. This process is condensation reaction. First, hydrazine(N2H4) is
heated with the 3-nitrophthalic acid. This reaction is most conveniently carried
out in the high-boiling solvent triethylene glycol. To boil off excess water, the
temperature is raised to the point where the dehydration occur.
Within a few minute, this process is completed.
Luminol(C8H7O3N3) powder is mixed with a liquid containing hydrogen peroxide(H2O2), a hydroxide (OH-). In order to produce a strong glow, they need a catalyst to accelerate the process. The catalyst in this case is the iron in hemoglobin . If hemoglobin and the luminol mixture come in contact, the iron in the hemoglobin accelerates a reaction between the hydrogen peroxide and the luminol. In this oxidation reaction, the luminol loses nitrogen and hydrogen atoms and gains oxygen atoms, resulting in a compound called 3-aminophthalate.
