Nitrate broth에 세균을 접종한 후, 37도 에서 24시간동안 배양한다. 배양 후,
Reagent A : 5g alpha-naphthylamine litre 5N acetic acid
Reagent B : 8g sulphanilic acid in 1 litre 5N acetic acid. 를 첨가한다.
각각의 시약을 다섯방울 넣고 흔든 뒤 1-2분 후에 결과를 관찰한다.
