SDS와 NaOH를 이용해서 cell을 깨고 chromosomal DNA와 protein등을 denaturation 시킴
High pH : DNA가 base pairing을 못함
Plasmid의 경우 작고 CCC form 이기 때문에 완전히 나눠지지 못함
pH가 정상으로 돌아오면, chromosomal DNA는 Protein등와 함께 침전되고 Plasmid는 상층액에 속함
